چکیده

I read with interest the recent article of Navarro-Locsin et al from the Philippines on the local rate of BRAF mutation in thyroid cancer. BRAF is a key genetic event in the initiation and progression of papillary thyroid carcinoma (PTC). It is a well-established diagnostic marker useful in the preoperative evaluation of indeterminate thyroid nodules. In addition, BRAF mutation attracts substantial interest as a therapeutic target and prognostic tool. It is believed that knowing the local BRAF mutation rate is of importance not only to the particular patient but also to health care providers. BRAF testing alone (opposite to multi-target gene panels) is deemed to be more cost-effective in regions with high prevalence of BRAF mutation. A rate of BRAF in PTC was extensively reported from Western countries (USA and Europe), with the consistent prevalence of 40-45% across different regions. Summary on BRAF mutation rate in Asian countries is shown in Fig. 1. Available data from South and Southeast Asia are very

A Review of Driver Genetic Alterations in Thyroid Cancers

Thyroid cancer is a frequent endocrine related malignancy with continuous increasing incidence. There has been moving development in understanding its molecular pathogenesis recently mainly through the explanation of the original role of several key signaling pathways and related molecular distributors. Central to these mechanisms are the genetic and epigenetic alterations in these pathways, su...

Targeting of the BRAF gene in papillary thyroid carcinoma (review).

Papillary thyroid carcinoma (PTC) is the most common type of thyroid malignancy and includes several histological variants. Patients suffering from PTC usually have a good outcome. However, a group of PTC patients still encounter high rates of recurrence and mortality. The BRAFV600E mutation is the most common genetic alteration identified in PTC, ranging from 29 to 83%, with the average rate a...

Tumorigenesis of Papillary Thyroid Cancer Is Not BRAF-Dependent in Patients with Acromegaly

INTRODUCTION Several studies have reported a high frequency of papillary thyroid cancer (PTC) in patients with acromegaly. The aim of this study was to determine the prevalence and predictors of thyroid cancer in patients with acromegaly and to investigate the frequency of the BRAFV600E mutation in PTC patients with and without acromegaly. MATERIALS AND METHODS We conducted a retrospective st...

Clinical implications of the BRAF mutation in papillary thyroid carcinoma and chronic lymphocytic thyroiditis

BACKGROUND The purpose of this study was to examine the possible prognostics and clinicopathologic characteristics underlying the BRAFV600E mutation and papillary thyroid carcinoma (PTC) coexisting or in absence of chronic lymphocytic thyroiditis (CLT). METHODS This study was conducted on 172 patients who had undergone total thyroidectomy or unilateral total thyroidectomy for PTC; the patient...
